Yes you can but I request you to elaborate the requirement.
I want exclude those folders on On Access Scan.
I had a problem last weekend when my developers were doing a test.
Can I create a exclusion on ON Access Scan for this situation?
Well I'll not recommend to do this exclusion as process details (.exe) are missing here which will access these 2 folders for the developers. I'll suggest to run McAfee profiler on his/her machine when developer do his activity, where it will give you correct scan count to particular folder and process. based on results you can add into LOW risk policy.
A quick question, Are you using low and high risk or only default? Are the Machines servers or workstation?
It should not be problema creating those exclusions under OAS.

I am using only default in workstations. I don´t have those kind of exclusions in our Servers.
If you are using only default than by adding folder exclusion will work for you.
C:\Users ----- Not recommended to add this, better try to get exact folder instance under Users folder and add it to exclusion.
C:\ProgramData\HP ---- Can be add to exclusion
