Do you have VSE installed on the machine? If yes then please look at Access protection log. This log will help in isolating whether port 25 is blocked or not.

Moved to ePO for better attention.
In the ePO server, system tree=>Assigned policy=>select the VSE version.
Select either Workstation or server depending on the nature of the machine and modify the Access protection policy.
