No special requirements, but I would let ePO make it's own instance of Sql or install a fresh instance of Sql for ePO to use rather than share the WSUS copy.
How to do that is all in the installation guide.
If you try to install ePO on a server already runing WSUS using internal DB, ePO-Instalation fails cause it can't install SQL Express 2005.
See this note in the installation guide (Page 8):
Using ePolicy Orchestrator with a database
A database must be installed before ePolicy Orchestrator can be installed. Any of the following
databases, if previously installed, meets this requirement.
• SQL Server 2005
• SQL 2005 Express
• SQL 2008
• SQL 2008 Express
SQL 2000 is not supported.
If none of those databases was previously installed, the ePO installation wizard detects that no
database is present and offers you the opportunity to install SQL Server 2005 Express.

Since WSUS installs an instance of Sql, the option to install Sql Express 2005 through ePO is disabled - this is by design.
You will need to install a separate instance of Sql or share the WSUS instance. I would suggest a separate installation for ease of maintenance.
Got it running after tring sever things with our SQL-aunt...
1. Download the SQL 2005 Express Install from MS
2. Install SQL Express 2005 regarding following article --> http://social.msdn.microsoft.com/Forums/en-US/sqlsetupandupgrade/thread/b906c398 -e40a-4f7c-ad0f-1b4ea07144af >>> last entrie
3. Activate the TCP-Port for the new DB created in step 2
4. Install ePO 4.5
5. Have FUN
