I run ePO 3.6.1 in VMware with no problems (well have a current issue but only just happened)
But ePO 4.0 runs ok on VMware. if you use esx 3.5 you need patch 4.

Will probably be ePO 4 but what needs to be patch 5 for ePO 3.5 ? - VMWare or ePO ?

I think what he was trying to say was that if you plan to install ePo 4.0 on a Virtual Machine running on VMware ESX 3.5, then you'll need to install ePO 4.0 patch 4 or later.

We have epo 4.0 patch 4 on Win2003 x64 Virtual Server R2 and it runs without problems. Currently we are testing epo 4.5 on Win 2008 x64 Hyper V and it works fine at the moment.
Yep, I have ePO 4 patch 4 installed on a Server 2003 Sp2 Virtual Machine running in VMWare Server 2.0.1
