You should be able to disable MA Self Protection through the registry - forgot which key, you will need to look around.  Yet, depending on your MA & VSE/ENS policy; you may not be able to edit the key or if edited it may toggle back on - this is all driven by MA & VSE/ENS policy through ePO.

I need to modify (LockdownStatus)  Registry value on below registry to disable MA Self Protection.

HKEY_LOCAL_MACHINE\SYSTEM\ControlSet001\Services\macmnsvc

HKEY_LOCAL_MACHINE\SYSTEM\ControlSet001\Services\masvc

HKEY_LOCAL_MACHINE\SYSTEM\ControlSet001\Services\McAfeeFramework

HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Services\macmnsvc

HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Services\masvc

HKEY_LOCAL_MACHINE\SYSTEM\CurrentControlSet\Services\McAfeeFramework

But unable to modify the above registry value. I have tried to change the Ownership of the Registry but getting access Denied.

I am the ePO administrator. I have ePO test and I was trying to disable the MA self protection to change "server sitelist.xlm" file on a single system.

Double check your Access Protection - you may need to disable Access Protection to modify the registry.
