In server settings where you configure your email server, you can send a test email to the new recipients from there without affecting any other users.

Since you are going to test, disable the existing automatic response which will ensure the current recipents won't be notified.
Create a new automaticresponse which will be duplicate of the previous one and add the new reciepent and delete the reciepent you dont want to notifiy. Once test is successful, disable the new automatic response and enable the previous one.
