We have been asked to block our Internal SharePoint site by a client for a specific location. We do have an existing URL filtering per location, but how do we block an internal website, if there is a rule to bypass internal traffic?

In order to create a rule for a specific location/proxy ,You can add criteria to your rule sets that would be location specific. For example:

Look for How do I create a region specific policy? part in above link.
You can then create a rule inside the rule set to block your sharepoint site.
Your sharepoint site for example is https://abc.com, then you can create a rule URL.host matches *abc.com* and set action as Block. This is just an example reference.
This blocking rule should be above your bypass rule.
