Your first weapon would be to initiate System Restore to before all this started. If you can't do it in regular mode simply power off and reboot into Safe Mode and try starting it there.
Second weapon would be to try Stinger and Malwarebytes Free, both linked in the last link in my signature below.
They can be downloaded to your desktop if necessary in Safe Mode with Networking and if you "Save as" and change their names in the process the malware shouldn't prevent them being run in regular mode.
However Malwarebytes Free can also be installed, updated and run in Safe Mode with Networking if necessary.

(They do have one for Vista Defender 2013 however).
